William "Isler" is William Iles Jr. his bondsman, Jacob Overholser, is his brother-in-law, husand of Hannah Iles.
The best way to find people in east Tennessee before about 1830 is through tax lists. I don't have the book title handy, but William Iles and William Kerlin are on the Washington County tax lists in the same company from 1814 to 1819. William Iles first appears on an 1801 list. I apparently haven't checked the tax lists from the 1820s. William Kerlin moved to Indiana in the 1830s.
The birth in Scotland comes from the Kerlin family tradition, which says William and Jane Iles were from Scotland.
I haven't found Lewis Barnet, either, other than the marriage record.
